According to Pfizer, immediately after application, Depo-Provera plasma concentration reaches 1.7 nanomole per liter and falls to that initial level after 12 weeks. NHS Choices states there is no definitive agreement about when the drug completely leaves the system; ovulation is inhibited up to five and half months after application.

Pfizer states that Depo-Provera is a long-acting progestational steroid, containing medroxyprogesterone acetate. According to Pharmacia and Upjohn Company, the drug has a terminal half-life of 43 days and reaches the maximum serum concentration after 8.8 weeks. Plasma half-life of the drug is about 6 weeks and is eliminated from the body through urine and feces.
